Rabeprazole + Domperidone

Indications

Rabeprazole + Domperidone is used for: Short-term treatment of GERD.

Adult Dose

Adult & adolescent >12 yr 1 cap once daily for 4-8 wk.

Child Dose

Renal Dose

Administration

Should be taken on an empty stomach: Take 15-30 min before meals. Swallow whole, do not chew/crush.

Contra Indications

Hypersensitivity to substituted benzimidazoles. Prolactinoma, severe hepatic impairment, GI hemorrhage, mechanical obstruction or perforation.

Precautions

Exclude gastric malignancy prior to treatment. Prolonged treatment. Pregnancy & lactation. Childn. Elderly.

Interactions

Monitor for increase in INR & prothrombin time when given w/ warfarin, cyclosporine, ketoconazole, ritonavir, erythromycin.

Adverse Effects

Side effects of Rabeprazole + Domperidone : Asthenia, fever, allergic reaction, chills, malaise, substernal chest pain, neck rigidity, photosensitivity reaction. CV, GI, endocrine, hemic & lymphatic, metabolic & nutritional, musculoskeletal, CNS, resp, urogenital, skin & special senses disturbances.

Mechanism of Action

Rabeprazole is a PPI that suppresses gastric acid secretion by inhibiting H+/K+ ATPase at the secretory surface of the gastric parietal cell. Domperidone is a peripheral dopamine-receptor blocker. It increases oesophageal peristalsis, enhances gastroduodenal coordination and lowers oesophageal sphincter pressure, gastric motility and peristalsis, thus facilitating gastric emptying and decreasing small bowel transit time.
